CLOTAZOLE-OD is Selective Phosphodiesterase III Enzyme Inhibitor, quinolinone derivative, Antiplatelet Agent(Platelet Aggregation Inhibitor) The drug exerts it`s antithrombotic action by interfering with platelet aggregation. It inhibits enzyme phosphodiesterase III and causes an increase in cyclic AMP in platelets and blood vessels which results in inhibition of platelet aggregation and vasodilatation. It reversibly inhibits platelet aggregation induced by stimuli; such as thrombin, ADP, collagen, arachidonic acid, epinephrine, stress, etc. The drug also reduces triglyceride level increases HDL cholesterol level. Absorption: Absorbed orally and bioavailability increases when administered along with a fatty meal. Distribution: Widely distributed in a protein bound form, Metabolism: Extensively metabolized in liver in to active metabolites. Excretion: Excreted mainly through urine as metabolites (about 74%), and a portion (about 20%) is excreted through faeces.
